Hs U2OS Synchronized Cell Samples Velos U2OS cells were grown in DMEM medium supplemented with 10% bovine serum, harvested by trypsinization, washed twice in phosphate-buffered saline (PBS), resuspended in 0.5 ml PBS, and lysed by adding 0.5 ml of 10 mM Tris pH 7.5, 10 M urea, 0.1% Rapigest. Dm_Kc_lysate_exp_078 1.Collect cells, wash with PBS.2.lyse cell with buffer D? After centrifugation, the supernatant is diluted with buffer D (buffer D? without Triton) to 0.75% Triton, centrifuge again. 3.Add 100ul anti-HA beads to the supernatant, shake at 4C for 4 hrs.4.Wash beads with buffer D+0.5% Trion X-100 4 times, then with buffer D (without protease inhibitor) twice. Bound proteins are eluted by 0.2mM glycine (pH 2.5) followed by precipitation with acetone.
